Bacterial 16S + fungal ITS NGS control plasmid
This plasmid is designed to function as a “spike-in” recovery control for Next Generation Sequencing sample prep and library construction for both bacterial 16S and fungal ITS quantitative community composition projects. The plasmid contains synthetic, not-present-in-nature, bacterial 16S sequence and fungal ITS region sequence, while retaining the commonly used Earth Microbiome Project barcoding primer sequences (bacterial 16S; fungal ITS). The synthetic 16S and ITS sequences are identical in length and average G/C content to wild-type sequences. By adding a small but accurately quantified number of plasmid molecules to a microbiome sample, the plasmid can serve as recovery and amplification control, since reads from the synthetic plasmid sequences are easily distinguished from reads from natural sequences present in a sample. The linearized plasmid is available in a 1 x 104 copies/µL dilution (the normal spike-in concentration), as well as 10-fold higher and lower (1 x 105 copies/µL and 1 x 103 copies/µL).
